Efficient Cloning of Taq-Amplified PCR Products

TA Cloning® Kits are designed for cloning Taq-amplified PCR products directly from a PCR reaction using an overnight ligation step. The kits use a pCR® vector and yield >=80% recombinants with >=90% of the recombinants containing inserts. Some of the convenient features of the pCR® vectors are:

• 3´-T overhangs for direct ligation of Taq-amplified PCR products
• Choice of T7 (pCR®2.1) or T7 and Sp6 (pCR®II) promoters for in vitro RNA transcription and sequencing
• Versatile polylinker with flanking EcoR I sites for easy excision of inserts
• Choice of kanamycin or ampicillin selection
• Easy blue/white colony screening
• M13 forward and reverse primer sites for sequencing
  
  Contents and Storage:
  TA Cloning® Kits contain linearized pCR®2.1 or pCR®II vector, T4 DNA ligase, 10X ligation buffer, dNTPs, 10X PCR buffer, sterile water, and controls. Competent cell kits contain One Shot® Chemically Competent E. coli, S.O.C. medium, and a supercoiled control plasmid. Store One Shot® E. coli at -80°C. Store all other components at -20°C. All reagents are guaranteed stable for 6 months when properly stored.
